Abstract

We aimed to identify biological cell processes involved in experimental right ventricular failure (RVF) due to chronic pressure overload. Wistar rats were subjected to pulmonary artery banding (PAB) (1.1mm, n=9) or sham surgery (CON, n=7) and then daily checked for clinical signs of RVF (inactivity, ruffled fur, dyspnea, ascites). At the onset of clinical RVF, termination occurred coupled with rat without symptoms of RVF. Gene micro array was performed on right ventricle tissue using GeneChip Rat Gene 1.1 ST (Affymetrics). Quality control and normalization were performed according to international standards. Micro array data were analyzed using Database for Annotation, Vizualisation and Integrated Discovery (DAVID), Gene Set Enrichement Analysis (GSEA) and ErmineJ. The data indentified processes as result of PAB. The data also indentified processes involved in RVF. This study observed a not earlier mentioned upregulation of inflammation and coagulation in response of the chronic pressure overload and a downregulation of energy metabolism which contributes to failure.
