Abstract

Objective: The aim of this study was to analyze the in-vitro properties (cytotoxic and bacteriostatic effect) of commonly used antiseptics and their respective dilutions. We will use this data to discover a possible candidate for intra-operative wound irrigation. The final goal is to reduce post-operative infections via this method. Methods: Bactericidal effect of antiseptics was investigated via standard quantitative suspension test; effect was investigated on S. Aureus and S. Epidermidis. Cytotoxic effect was investigated on human fibroblasts and mesenchymal stem cells with cell viability measured via the WST-1 metabolism assay. Results: Povidone-iodine was the only antiseptic which possessed a concentration with bactericidal capabilities against both strains of bacteria whilst there was still some cell vitality left. Chlorhexidine could not be investigated properly on a monolayer of cells due to the inability to be mixed with standard solution compounds (PBS, physiological saline). The other tested antiseptics did not possess this capability. Conclusions: Povidone-iodine is the only antiseptic with full bactericidal capabilities whilst retaining some viability of cells. Therefore povidone-iodine in a concentration of 1.4 g/L is in this investigation the best option to use as on intra-operative irrigation antiseptic.
