Abstract

Background Organohalogen compounds, i.e. polychlorinated biphenyls (PCBs), are widely spread environmental pollutants, known to be neurotoxic for the developing brain. Hydroxylated metabolites of PCBs (OH-PCBs) seem to be even more toxic. However, little knowledge exists on the effects on human health. Objectives To determine whether prenatal background exposure to OHCs has an effect on the mental and motor development in 18-month-old infants. Methods We included 181 infants of two observational cohorts in the northern part of the Netherlands. We measured ten PCB and six OH-PCB congeners in maternal blood in the first cohort. In the second cohort, we measured PCB-153, three OH-PCBs, five 2,2’,4,4’-tetrabromodiphenyl ethers (BDEs) and three other OHCs in maternal blood. We used Bayley’s Scale of Infant Development II (BSID-II) to assess infant’s mental (MDI) en motor development (PDI). Results After adjusting for confounders, we found several associations. Higher exposure to 4-OH-PCB-146 (OR=1.160; p<0.001) and 4-OH-PCB-187 (OR=1.080; p=0.043) was associated with lower scores on MDI. Moreover, 4-OH-PCB-107 (OR=1.453; p=0.020), PCB-153 (OR=1.231; p=0.023), 2,2′-bis-(4 chlorophenyl)-1,1′-dichloroethene (p,p’-DDE) (OR=1.097; p=0.010) and BDE-154 (OR=1.257; p=0.010) measured in one cohort, also showed adverse associations with MDI scores. In the other cohort, exposure to 4-OH-PCB-107 was positively associated with PDI scores (OR=0.882; p=0.048). Conclusion Our findings indicate that at the age of 18 months, prenatal exposure to OHCs seem to have an adverse effect on mental development, whereas only 4-OH-PCB-107 seem to have a positive effect on motor development.
