Abstract
This paper investigates whether a selection of single nucleotide polymorphisms (SNP) of the oxytocin receptor (OXTR) gene moderate the oxytocin (OXT) effect on social orientation in adult males with- and without ASS. Social orientation was measured by looking at a neurophysiological response, the late long lasting parietal positivity (LPP), to emphaty-evoking pictures selected from the International Affective Picture System (IAPS). These pictures depicted pleasant, unpleasant and neutral scenes with and without human beings. In this randomized double-blind placebo-controlled crossover trial, subjects looked at these pictures after having nasally been administered either oxytocin or a placebo. The following OXTR SNPs were looked at: rs53576, rs2254298, rs2228485, rs237887, rs237897, rs13316193 and rs237889. The results showed that the OXTR SNPs rs2254298 and rs2228485 moderate the OXT effect on social orientation. The OXTR SNP rs237897 impacted the OXT effect on the orientation response to affective pictures of human beings in especially pleasant situations and this impact differed between the control group and the ASS group. A relationship was found between the OXTR SNPs rs2254298 and rs2228485 and self-reported measures of empathy. Our results suggest that gen varieties of the OXTR gene moderate the effect of nasally administered OXT on social orientation.
